Stam was born in Kincardine, Ontario, and grew up on a farm alongside her six brothers. As a child, her family was very religious, practicing the Christian faith. She originally wanted to be a dentist.

Stam was discovered in a local Tim Hortons coffee shop by Michèle Miller, an agent at the International Model Management agency in Barrie, Ontario, who found Stam on the way back from Canada's Wonderland, a theme park just outside Toronto.

Stam attended St. Joseph's High School in Barrie, Ontario, where she lived with roommate and fellow model Michelle V.

As soon as photographer Steven Meisel discovered the then 16-year-old Stam, he cast her in every ad campaign of his, and suddenly she was a star. "I guess I'm his muse," she has said of her mentor. Stam won the Los Angeles Model Look Search in 2002.

Her career was set off when she appeared on the cover of UK & German Vogue and she has appeared in advertisements for Marc Jacobs, Anna Sui, Vera Wang, Valentino, Miu Miu, Gucci, Prada, Dolce & Gabbana, Versace and Holt Renfrew. In 2004, Stam appeared in the short film Agent Orange.

Good friend and fashion designer Marc Jacobs recently created a bag inspired by and named after her (The Marc Jacobs Stam). Fashion Show, in which she had her own featurette and opened a Theme entitled "Pink".

For Spring 2006 Ready-to-Wear, Stam walked a total of 64 shows combined during the New York, Milan, and Paris Fashion Weeks. During Paris's Spring/Summer 2007 Haute Couture fashion week she walked for Chanel, Christian Lacroix, Christian Dior, Givenchy, and Jean Paul Gaultier. She currently stands at number five on the "Top 50 Models" rating at models.com.

In the May 2007 issue of American Vogue she was featured on the cover with fellow models: Doutzen Kroes, Caroline Trentini, Raquel Zimmermann, Sasha Pivovarova, Agyness Deyn, Coco Rocha, Hilary Rhoda, Chanel Iman, and Lily Donaldson as the new crop of supermodels.

In July 2007, earning at an estimated total of $1.5 million in the past 12 months, Forbes named her fifteenth in the list of the World's 15 Top-Earning Supermodels.

Currently she stars in campaigns for Bulgari, Lanvin and Escada. She also appears in the F/W 2007 Christian Dior, DKNY, Miss Sixty and Roberto Cavalli ad campaigns.

Stam's main trademark is her eyes, which are a light blue bordering on grey. She is also one of the models considered a "doll face." Others in this category include Gemma Ward, Lily Cole, Caroline Trentini, Lisa Cant, Devon Aoki and Heather Marks.

Stam previously dated Anthony Kiedis of the Red Hot Chili Peppers. In January of 2006 she was photographed topless while frolicking with Kiedis on the Caribbean island of St. Barts. On January 12, 2006 she was photographed with him during a Los Angeles Lakers/Cleveland Cavaliers basketball game at the Staples Center in Los Angeles. Before Kiedis, Stam dated Harrison Ford's son, Malcolm. Recently, in 2007, she split up with boyfriend Adam Goldstein.
