An EasyJet spokeswoman said crew found "a syringe hidden in a bin, covered in blood" in the plane's toilet.
Spanish Guardia Civil met Doherty and four friends but found no suspicious substances in the lavatories.
They were not arrested and were allowed to leave. Doherty's band Babyshambles are in the city to play at a festival.
The EasyJet spokeswoman said the crew became suspicious after Doherty spent "an unusual amount of time in the toilet".
'Agitated and aggressive'
The singer and his bandmates were travelling on flight EZY5135 from London Gatwick.
After the discovery of the syringe, Doherty was questioned by crew and became "agitated and aggressive", the spokeswoman said.
The captain contacted Spanish police, who met the plane, which was held on the ground for about half an hour.
The band have now been banned from travelling on EasyJet flights in the future, the spokeswoman added.
Babyshambles are due to perform at the Primavera Sound Festival in Barcelona on Thursday.

As I heard the report, his bandmates became concerned when he took longer then usual in the restroom. They reported it to the crew, who immediately performed a search after he left.
Of course his bandmates knew 'bloody well' what he was up too. They are probably hoping to get rid of him.

correct me if i'm wrong, but flights originating in countries BESIDES the US are not quite as stringent?
and truth be told, even in the US they dont catch a lot of things. there are ways to get on a plane with something if you want to bad enough.
